Induction of Broadly Cross-Reactive Cytotoxic T Cells Recognizing an HIV-1 Envelope Determinant

Abstract: An immunodominant determinant for cytotoxic T lymphocytes (CTLs) exists in the hypervariable portion of human immunodeficiency virus-1 (HIV-1) gp160. Three mouse CTL lines (specific for isolates MN, RF, and IIIB) were examined for recognition of homologous determinants from distinct isolates. Only MN-elicited CTLs showed extensive interisolate cross-reactivity. Residue 325 played a critical role in specificity, with MN-elicited CTLs responding to peptides with an aromatic or cyclic residue and IIIB-induced cel… Show more

“…In this report we compared the structures of a 15-residue peptide from the HIV-1IIIB V3 loop and a glycosylated analogue with N-acetylgalactosamine (NAG) c~-linked to the threonine four residues to the C-terminal side of the so-called immunodominant tip [18,19] (sequence -GPGR-) of the loop. As this peptide is also an immunodominant epitope for CTL recognition [20][21][22][23], we have also tested these peptides for their ability to be recognized by class I-MHC molecule-restricted CTL as measured by lysis of a tumor cell line incubated with the peptides.…”

“…We performed these studies using plasmid DNA encoding the HIV-1 gp120 protein as the immunogen. In BALB/c mice, CD8 + T cells recognize the immunodominant 10-amino acid p18sequence of the HXB2 gp120 protein [28]. We first explored the potential adjuvant effects of PEI-DNA complexes delivered by the intramuscular route.…”

“…Groups of naive recipient mice (n ϭ 4/group) received 500 l of purified IgG by tail vein injection before immunization with 10 8 vp of rAd5-Env. Env-specific CD8 ϩ T lymphocyte responses against the dominant P18 epitope (RGPGRAFVTI) (26) were assessed by D d /P18 tetramer binding assays (27,28). As shown in Fig.…”
