Induction of Cell Death in Renal Cell Carcinoma With Combination of D-Fraction and Vitamin C

Abstract: The present study demonstrates that the combination of PDF and VC can become highly cytotoxic, inducing severe cell death in ACHN cells. This cytotoxic mechanism appears to be primarily attributed to oxidative stress, accompanied by a G1 cell cycle arrest. Such cell death induced by PDF + VC could be more likely linked to apoptosis, as indicated by the modulation of apoptosis regulators (Bcl2, Bax, and PARP). Therefore, as PDF and VC may work synergistically to induce apoptotic cell death, they may have clinic… Show more

“…On the contrary, vitamin C assisted low-dose MTX to exhibit a strong cytotoxic effect in Hep3B cells. Similarly, recent studies also indicated that vitamin C can enhance anticancer agents to exert a strong cytotoxic effect on cancer cells, although the mechanisms remain unknown (48,7072). Thus, MTX-induced increases in H 2 O 2 levels may be one of the factors resulting in cytotoxicity noted in MTX-treated Hep3B cells.…”

“…Alternatively, there have been many reports on the antioxidant activities of vitamin C (4347). Moreover, some studies have demonstrated that vitamin C can exert anticancer activities in various cancer cells (4852). The present study demonstrated that vitamin C can diminish MTX-induced increases in H 2 O 2 levels.…”

“…Chen et al (23) showed as well that VHL-deficient 769-P renal cancer cells are sensitive to megadose Vc, but the authors did not assess whether VHL re-expression reduces this effect. Moreover, Alexander et al (65) demonstrated that ACHN renal cancer cells are highly sensitive to a combination of the D-fraction of the Maitake mushroom and Vc , although it should be noted that this cancer cell line is VHL-competent. VHL-defective renal cancers compose the vast majority of renal cancers in the adult (42), and a proportion of these cancers is familial in the context of the VHL syndrome (42).…”

“…As mentioned previously, the antitumor properties of the Maitake DFraction are attributed to its immunomodulatory effect. Reports have also indicated that Maitake directly induce apoptosis in tumour cells such as prostate [33], stomach [34], kidney [35] and breast [36] cancer cells.…”
