Induction of Fibrosis and Autophagy in Kidney Cells by Vinyl Chloride

Abstract: Vinyl chloride (VC) is a noninfective occupational risk factor. It is found in industrial chemicals, volatile organic compounds, cigarette smoke ingredients, etc. It is a kind of toxic gas that causes many diseases. VC exposure causes an increased risk of liver fibrosis and can result in angiosarcoma of the liver. Previous studies have shown that high-doses of VC exposure in mice resulted in acute death with marked tubular necrosis of the renal cortex. In this study, we assessed the nephrotoxicity of VC in vit… Show more

“…Additionally, PAI-1 is considered an autophagy activator in immune cells [53]. In a model of vinyl-chloride-induced renal fibrosis, activation of autophagy corresponded to increased PAI-1 expression, while expression of PAI-1 gene in HK-2 cell culture was increased under beclin-1 siRNA exposure [54]. Nevertheless, in our study we failed to find any relationship between plasma PAI-1 levels and autophagy parameters.…”

“…In addition, microplastics and nanoplastics elicit immunological responses [115,123,124], alter gene expression [88,103,113,114,125] and induce genotoxicity [113,126]. In kidney cells, VC stimulates the expression of fibrosis-related proteins, such as CTGF, PAI-1, and collagen 1, and autophagy-related proteins, such as Beclin 1 and LC3-II [127]. VC is also a carcinogenic factor and results in angiosarcoma of the liver [94][95][96].…”
