Induction of increased calcium uptake in mouse T lymphocytes by concanavalin A and its modulation by cyclic nucleotides

Abstract: The binding of concanavqlin A to T but not B mouse spleen lymphocytes increases Ca-2+ uptake in these cells which is measurable by 45 s and complete by 1 min. Dibutyrl cyclic AMP, but not sodium azide inhibits induced Ca-2+ uptake, wheras dibutyryl cyclic GMP enhances it. B cell mitogens do not cause a similar Ca-2+ uptake in mouse B lymphocytes. The induction of increased Ca-2+ uptake by T cells is discussed in terms of gated membrane channels for Ca-2+.

“…In mouse T lymphocytes, 'gated channels' for Ca", which could be opened directly by Con A binding to membrane glycoproteins, have been postulated, the operation of which may be regulated by cyclic nucleotides (18). From these results, combined with the present data, co-interaction between protein kinase A and protein kinase C in platelets is suggested to be involved in the inhibitory effect of Con A on 5-HT transport.…”

“…In addition, the influence of temperature, pH, a metabolic inhibitor, and potential competitive inhibitors was examined. The influence of temperature was examined by comparing antibiotic uptake at 4 and 37°C. The pH profile of CLDM uptake was measured in media that had been preadjusted to different external pH values (from 5.0 to 8.5) by the addition of 10 N HCl or 10 N NaOH.…”

“…Thus, the ACTH action to increase tissue cAMP concentration in both adrenal cortex and adipose tissue is calcium dependent (35)(36)(37). Further, in lymphoid cells in vitro, the mitogen concanavalin A induces calcium-dependent increases in the uptake of both 3-0-MG and glucose (38,39).…”
