Induction of mitochondrial apoptotic pathway by raloxifene and estrogen in human endometrial stromal ThESC cell line

Abstract: IntroductionEndometrial hyperplasia is a condition that occurs as a result of hormonal imbalance between estrogen and progesterone. Morphological disturbance of endometrial cells occurs consequently leading towards endometrial cancer. In therapy of endometrial hyperplasia SERMs are used to supress effects of locally high estrogen level in uterus. There is strong evidence suggesting that estrogen could be involved in cell death – apoptosis. There are no experimental data demstrating the direct apoptotic effect … Show more

“…To determine the effects of the combinations of investigated substances on the viability of ThESC cells and to compare them to the cytotoxic effects of their single drug application, the percentage of cytotoxic cells was determined by MTT assay following the 24‐hr treatment with the single dose of raloxifene (10 −9 m ), estrogen (10 −4 m ), methotrexate (10 −6 m ), and with the combination of raloxifene (10 −9 m ) either with estrogen (10 −4 m ) or with methotrexate (10 −6 m ). The particular concentrations of the substances were used based on the lowest exerted cytotoxic effects of these concentrations of drugs in ThESC cells, as shown in our previous study . The single drug's cytotoxic effects of investigated substances were clearly distinguished and did not exceed 10% of cytotoxic cells, as shown earlier .…”
