Diabetes mellitus (DM) during pregnancy is one of the leading causes of perinatal morbidity and birth defects. The mechanism by which maternal hyperglycemia, the major teratogenic factor, induces embryonic malformations remains unclear. Advanced glycation end products (AGEs) are known to accumulate during the course of DM and contribute to the development of diabetic complications. Employing a diabetic rabbit model, we investigated the influence of maternal hyperglycemia during the preimplantation period on AGE formation (pentosidine, argpyrimidine, and N 3 -carboxymethyllysine (CML)) in the reproductive tract and the embryo itself. As a consequence of type 1 DM, the AGE levels in blood plasma increased up to 50%, correlating closely with an AGE accumulation in the endometrium of diabetic females. Embryos from diabetic mothers had increased protein-bound CML levels and showed enhanced fluorescent signals for AGE-specific fluorescence in the blastocyst cavity fluid (BCF). The quantification of CML by HPLC-mass spectrometry (MS/MS) showed a higher amount of soluble CML in the BCF of blastocysts from diabetic rabbits (0.26G0.05 mmol/l) compared with controls (0.18G0.02 mmol/l). The high amount of AGEs in blastocysts from diabetic mothers correlates positively with an increased AGER (receptor for AGE (RAGE)) mRNA expression. Our study gives alarming insights into the consequences of poorly controlled maternal diabetes for AGE formation in the embryo. Maternal hyperglycemia during the preimplantation period is correlated with an increase in AGE formation in the uterine environment and the embryo itself. This may influence the development of the embryo through increased AGE-mediated cellular stress by RAGEs.

Advanced glycation endproducts (AGEs) is implicated in the pathogenesis of diabetic complications. Inhibiting the formation of AGEs and interfering with AGEs-mediated inflammation are two practicable strategies for developing a dietary adjuvant against diabetic complications. This study evaluated the protective capacities against diabetic complications of several spices based on their inhibition of the formation of AGEs in an in vitro BSA/glucose system and on the AGEs-induced production of proinflammatory cytokine in RAW 264.7 macrophages. Among the tested spices, cinnamon exhibited most strongly inhibited both the formation of AGEs and the AGEs-induced production of nitric oxide, interleukin-6 and tumor necrosis factor-α. Additionally, correlative results revealed that the capacity of spices to inhibit the formation of AGEs is attributable to phenolic compounds and, in contrast, the capacity to inhibit AGEs-induced inflammation is attributable to condensed tannin. This investigation demonstrates the potential of cinnamon to serve as a dietary adjuvant against diabetic complications.

Abstract. recurrence and metastasis are the major factors associated with the poor prognosis of non-small cell lung cancer (nsclc). It has been shown that multiple chemokines and their receptors are related to the progression and metastasis of nsclc. the aim of this study was to conduct an investigation into whether ccl21 and its receptor, ccr7, play a role in nsclc invasion and metastasis. We used Western blotting, immunocytochemistry and flow cytometry to detect CCR7 protein expression in four nsclc cell lines eKVX, Hop-62, ncI-H23 and slu-01; and we conducted a cell migration experiment to observe the pseudopodia formation and mobility of the lung cancer cells. the concentration of intracellular calcium was measured by fluorescence microscopy. ccr7 protein was positively expressed in the four nsclc cell lines eKVX, Hop-62, ncI-H23 and slu-01. Following ccl21 stimulation, obvious pseudopodia formation of lung cancer cells was observed. the cell migration experiment showed that following incubation with ccl21, the number of eKVX cells which passed through the polycarbonate micro-porous filter membranes also increased to an obvious extent. After ccl21 incubation, the intracellular ca 2+ level of the eKVX cells increased to an obvious extent. chemokine ccl21 facilitates the migration of lung cancer by changing the concentration of intracellular ca 2+ . the ccl21-ccr7 axis may play an important role in nsclc invasion and metastasis. It may also be a potential target for nsclc therapy or for prevention of the recurrence and metastasis of nsclc.
