“…In addition, while DNAm can cause changes in hormone production and hormone receptor expression, hormones can also cause changes in DNAm, for example, through the regulation of their associated enzymes (DNMTs; Baumbach & Zovkic, 2020). In barramundi specifically, estrogen treatment resulted in decreased DNAm and increased expression of cyp19a1a , as well as early male‐to‐female sex change (Banh et al, 2021). It remains unknown whether changes in DNAm occur prior to or following changes in gene expression and natural hormone production, which lead to sex change in fish.…”

“…To analyze DNAm levels of the region surrounding the start codon of cyp19a1a , esr1 , dmrt1 , and nr5a2 , a targeted bisulphite amplicon sequencing (BSAS) approach, adapted from Masser et al (2013), was applied. These regions were selected based on our previous work, both published ( cyp19a1a and nr5a2 ) and unpublished ( esr1 and dmrt1 ), to identify those showing significant correlations with phenotypic sex (Banh et al, 2021; Budd, 2020; Domingos et al, 2018). While the region surrounding the transcription start site is typically the most informative (Piferrer et al, 2019), not all sex‐related genes show sex‐specific DNAm patterns in amplicons near this region (e.g., sox9 ; Figure ).…”

“…Steroid hormones are natural and induce sex change processes [ 50 ], among which E2 is one of the most potent estrogens impacting sex differentiation [ 51 ] and MT is one of the most potent androgens impacting sex differentiation [ 52 ]. For instance, E2 induces sex reversal in P. sinensis and the protandrous barramundi ( Lates calcarifer ) [ 53 , 54 ], and MT controls secondary sexual characteristics in Siniperca chuatsi as well [ 55 ]. The same phenomenon has been confirmed in mammals and fish [ 56 ].…”

“…The experimental results suggest that estradiol treatment induced the upregulation of CsFOXL2. It indicates that estradiol may play a role in ovarian development by stimulating FOXL2 to induce estrogenic feedback mechanism (Narisato et al, 2006;Li et al, 2014;Banh Quyen et al, 2021). However, the expression level of CsFOXL2 was decreased at estradiol concentration of 50 µg/L compared with estradiol concentration of 5 µg/L.…”
