Induction of Specific Immunoglobulin A and Th2 Immune Responses to P6 Outer Membrane Protein of NontypeableHaemophilus influenzaein Middle Ear Mucosa by Intranasal Immunization

Abstract: Nontypeable Haemophilus influenzae (NTHI) is a major pathogen of otitis media. One of the outer membrane proteins of NTHI, P6, is an antigen common to all strains and is considered as a candidate for mucosal vaccine. To elucidate the possibility of developing a nasal vaccine against nontypeable Haemophilus influenzae (NTHI) and to investigate mucosal immune responses in the middle ear, mice were immunized intranasally with the P6 outer membrane protein of NTHI, and P6-specific immune responses in the middle ea… Show more

“…NALT-like structures are also present in young humans, but these disappear at an early age (10). Immune responses within the nasal sinuses provide IgA-dominated protection that carries over to the mucosa of the intestines and lower and higher respiratory cavities, as well as ocular tissues and auditory cavities (20,25,36). Furthermore, NALTs function as collection centers for sampling airborne antigens and debris from colonizing microorganisms (13,29,33).…”

“…Another set of COM mice was made, and MNCs from the middle ear were harvested from groups of 5 mice at each time point. For the isolation of MNCs from the MEM, we established a dissociation method (23). Briefly, the skin was removed carefully from the head region, and the bulla was isolated from the temporal bone.…”

“…CTB is a nontoxic binding moiety of cholera toxin; it is composed of a ring of five identical polypeptides that bind with high affinity to GM1 and other ganglioside cell surface receptors and promote the entry of the A subunit into the cell (47,50). Oral or intranasal immunization has been shown elsewhere to successfully induce protective immunity against a variety of viral, bacterial, and protozoan infections (29,31,39,42,49,61). More recently, the possibility of using CTB as a mucosal adjuvant in humans has been reported (8).…”
