Influence of pegylated interferon-α therapy on plasma levels of citrulline and arginine in melanoma patients

Abstract: The aim of this study was to evaluate the effect of pegylated interferon-alpha (PEG-IFN-alpha) on the plasma citrulline/arginine ratio, regarded as an index of nitric oxide (NO) synthesis, in patients with high-risk melanoma. Forty patients were randomly assigned to either PEG-IFN-alpha treatment (n = 22) or to observation only (control group, n = 18). The treatment group received 6 microg PEG-IFN-alpha/kg once a week during 8 weeks, followed by a maintenance dose of 3 microg/kg/wk. Blood was collected at diff… Show more

“…In this study, the ratio of the amino acids citrulline and arginine (Cit‐Arg ratio) was calculated as an index of NO synthesis. This method has been reported to be sufficiently accurate and reproducible, and it can be used as an effective index to reflect the NOS‐NO system activity . Furthermore, we selected a special subtype of depression (MDD) for study, using the same SSRI monotherapy.…”

“…In addition, the symptomatic and euthymic subgroup of BP-I patients on lithium monotherapy both demonstrated a lower Cit-Arg ratio as compared to healthy controls as well as to the two patient groups with depressive disorders. This ratio is regarded as an index of the NO synthesis [13] and/or NO synthase activity [12] and thus, NO formation may be endangered in bipolar patients.…”

“…The NO synthesis was approximated by calculation of the ratio between the plasma levels of the amino acids citrulline and arginine (Cit-Arg ratio). Both NO and citrulline are formed from arginine, a reaction catalyzed by the enzyme NO synthase, and the Cit-Arg ratio is regarded as an index of the NO synthesis [12,13] .…”

“…In this process tetrahydrobiopterin (BH4) is a necessary cofactor. We have shown that the ratio of the plasma concentrations of citrulline (Cit) and arginine (Arg), the so-called Cit/Arg ratio, can be regarded as an index of NO synthesis (Fekkes et al 2007). …”
