Influence of the difference of breastfeeding volume on a rat model of oxygen-induced retinopathy

Abstract: This study aimed to investigate the effects of abundant breast milk intake on rats model of oxygen-induced retinopathy (OIR). Neonatal Sprague–Dawley rats were randomly assigned to expand litters of 7 pups/litter (7-rats group) and 14 pups/litter (14-rats group). They were exposed to 80% oxygen from postnatal day (P) 0 to P12. Body weights were measured daily. At P13 and 18, rats were sacrificed, and the blood and eyes were collected. Retinal neovascularization (NV) score, total retinal area (TRA), avascular a… Show more

“…administered with 0.5 mL of Conbercept ophthalmic injection (1.0 mg/kg), different doses of SPN (5.0 and 10.0 mg/kg) or normal saline (NS). Five days later (on PD 17), the rat pups were sacrificed (the pups in the control group were bred under normoxia condition until PD 17), and the eyes were enucleated and fixed in fresh 4% paraformaldehyde for 2 h. Eyecups were dissected and the retinal flat mounts were created and stained with ADPase stain as described previously [54]. The retinal avascular areas were assessed through visualization of the retinal vasculature using infusion of ADPase staining examined under fluorescent microscopy.…”

“…Also, the intravitreal administration of anti-VEGF agent in an attempt to suppress the development of oxygen-induced retinopathy of prematurity has been studied, but there are some concerns of systemic effect of anti-VEGF agent which escaped from the eye into the systemic circulation. ( 23 ) In this study, VEGF levels in the aqueous humor did not change after 2-week administration of AX, at 6 mg/day, as a supplement. This dose corresponds the amount of AX in, for example, a 300 g salmon fillet, within the expected range obtained through a normal daily diet.…”

“…The dose selection of Conbercept ophthalmic injection is based on our previous studies. Five days later (on PD 17), the rat pups were sacrificed (the pups in the control group were bred under normoxia conditions until PD 17), and the eyes were enucleated and fixed in fresh 4% paraformaldehyde for 2 h. The eyecups were dissected and the retinal flatmounts were created and stained with ADPase 25. The retinal avascular areas were assessed through visualization of the retinal vasculature with infusion of ADPase stain examined under fluorescent microscopy.…”
