PURPOSE:To evaluate the influence of the cyclosporine in liver regeneration in rats submitted to an experimental model of 70% hepatectomy. METHODS:Forty male rats were randomly divided in four subgroups (C.24h, C.7d, E.24h, E.7d), according to the drug used and the day of sacrifice (24 hours and 7 days). Cyclosporine (10mg/Kg/day) was given to the study subgroup and 1 ml of 0.9% sodium chloride was to the control subgroup. Resection of left lateral lobe and median lobe performing 70% of liver mass. During the animals' death, KWON formula was applied. Counting of mitotic figures and percentage of positive nucleus with PCNA and Ki-67 were evaluated. RESULTS:In the 2 nd , 4 th PO and death days, E.7d lose more weight than C.7d. Regarding to the KWON formula, the C.7d regenerated more than the C.24h and the same with the E.7d. Comparing between the groups, only E7d subgroup was statistically significant compared with C.7d, showing the stimulating effect of cyclosporine in liver regeneration. Immunohistochemestry had significant results between the study subgroups. The mitotic index revealed statistical differences in the control subgroups. CONCLUSION:Cyclosporine, in spite of being an immunosuppressive drug, has a positive effect in liver regeneration, although reduce the animal's body weight.

The ligation of portal branches is known to induce an atrophy of the deprived lobes and a hypertrophy of the nonligated lobes. In this work, we examined if this response occurs when the ligation affects 90% of the liver mass and if it is able to influence the survival rate after a hepatectomy of this magnitude. In male Wistar rats, major portal branches were ligated, keeping open only a small branch supplying the papillary lobes. The ligated lobes underwent a progressive weight loss and their DNA synthesis remained at the level of the controls. The nonligated lobes started to gain weight on the first postoperative day and had increased about 5-fold by the 14th day. The DNA synthesis in these lobes was greatly enhanced with a peak increase at 24 h. Two weeks after a similar ligation of portal branches, the ligated lobes, representing initially 90% of the liver mass, were resected and 80% of the rats survived. In the control group submitted only to a sham operation 2 weeks before, all rats died within 48 h after resection of the corresponding lobes. These results demonstrate that ligation of portal branches feeding 90% of the liver mass can be well tolerated and induces an atrophy of the ligated lobes and a hypertrophy of the nonligated lobes. This procedure, which improves the survival rate after an extended hepatectomy performed 2 weeks later, could find some clinical application.

Augmentation of hepatocyte proliferation by immunosuppressant pretherapy is associated with up-regulation of malondialdehyde production

We studied the relationship between augmentation of liver regeneration with immunosuppressants and malondialdehyde (MDA, an end-product of lipid peroxides) production. MDA was determined using the thiobarbituric acid reaction. Rats underwent a 4-day treatment of FK506 (FK, 1 mg/kg per day), cyclosporine (Cs, 10 mg/kg) or azathioprine (AZA, 1 mg/kg) by gavage prior to 70% hepatectomy. They were then divided into four groups: (1) controls (vehicle-treated); (2) FK; (3) Cs; (4) AZA. MDA levels, uptake of BrdU (5-bromo-2-deoxyuridine) in the liver and serum biochemistry were investigated 24 h after hepatectomy. Immunosuppressant pretherapy significantly stimulated BrdU uptake by hepatocytes, in association with increased MDA production, while there were no differences in serum liver injury parameters among the groups given or not given immunosuppressants. The implications of the rising MDA values during liver regeneration are discussed with respect to immunosuppression and a measure of lipid peroxidation. Additional study indicated that one immunodepressant pretreatment (24 h prior to hepatectomy) was effective for up-regulation of liver regeneration.
