Influences of dietary n−3 long‐chain PUFA on body concentrations of 20∶5n−3, 22∶5n−3, and 22∶6n−3 in the larvae of a marine teleost fish from Australian waters, the striped trumpeter (Latris lineata)

Abstract: We determined the effect of dietary long-chain (> or = C20) PUFA (LC-PUFA), 20:5n-3 and 22:6n-3, on larval striped trumpeter (Latris lineata) biochemistry through early development and during live feeding with rotifers (Brachionus plicatilis). Rotifers were enriched using seven experimental emulsions formulated with increasing concentrations of n-3 LC-PUFA, mainly 20:5n-3 and 22:6n-3. Enriched rotifer n-3 LC-PUFA concentrations ranged from 10-30 mg/g dry matter. Enriched rotifers were fed to striped trumpeter … Show more

“…The ALA level was high in the intestine of sea cucumbers fed diet ALA 12.71 , followed by those fed diets ALA 7.46 and ALA 0 . The content of EPA and DHA in tissues increased with increasing dietary EPA and DHA, which is in consistent with the PUFA level reported in the muscle of C. striata and Latris lineate larvae before 18 days posthatching (Bransden et al, ; Jaya‐Ram, et al, ).…”

“…The dietary fatty acid data is limited by the lack of replication, and needs to be interpreted with caution. Nonetheless, there were strong relationships among dietary fatty acids and fatty acid composition of larvae in both experiments as has been demonstrated for striped trumpeter previously (Bransden et al, 2004) and other species (Mourente et al, 1993;Rodriguez et al, 1994;Koven et al, 2001). Rotifers enriched with AlgaMac, under comparable conditions in our laboratory, have been reported to have up to 20% greater total fatty acid but lower proportions of essential LC-PUFA.…”

“…An aliquot of total lipid was also transesterified with a methanol/chloroform/hydrochloric acid (10 : 1 : 1, by vol) solution to form fatty acid methyl esters (FAME). The extraction and methylation procedures have been validated for a range of small samples including microheterotrophs (Lewis et al, 2000), striped trumpeter larvae and rotifers (Bransden et al, 2004;unpublished data). FAME were extracted, concentrated and an internal injection standard (methyl tricosanoate) added, the latter for Experiment 2 animals only.…”

“…Despite recent advances, hatchery‐produced juveniles have exhibited a high incidence of malformations (Cobcroft et al 2001; Trotter et al 2001; Battaglene and Cobcroft 2007). Research has identified the required content of long‐chain fatty acids and selected vitamins in live feeds (Bransden et al 2004a, 2004b, 2005a, 2005b; Brown et al 2005a,b). Historically, striped trumpeter postlarvae were reared using enriched Artemia until 100 d posthatch (dph).…”
