Infusion of adipose‑derived mesenchymal stem cells inhibits skeletal muscle mitsugumin 53 elevation and thereby alleviates insulin resistance in type 2 diabetic rats

Abstract: It is widely accepted that infusion of mesenchymal stem cells (MSCs) ameliorates hyperglycemia by alleviating insulin resistance in rats with type 2 diabetes mellitus (T2D). However, the detailed underlying mechanisms are not clearly defined. Mitsugumin 53 (MG53) is an E3 ligase that has recently been implicated in the aggravation of insulin resistance by promoting the ubiquitinoylation of insulin receptor substrate‑1 (IRS‑1) in skeletal muscles. It was therefore hypothesized that MG53 may be involved in MSC‑m… Show more

“…Adipose-derived MSCs were isolated and purified from the immature rats as previously described (20). Briefly, the rats were anesthetized with pentobarbital sodium intraperitoneally (60 mg/kg) and sacrificed by cervical dislocation.…”

“…After that, cells were washed twice and cultured in a low glucose DMEM (Gibco, Gran Island, NY, USA) supplemented with 10% fetal bovine serum (Gibco, Gran Island, NY, USA), penicillin (80 units/ml), and streptomycin (0.2 mg/ml). MSCs were identified as previously described (20). Immunologic phenotypes of third passage (P3) MSCs were analyzed by flow cytometry.…”

“…As described previously (22,23), MSCs derived from fat were isolated and purified from immature rats. A total of 3 rats were euthanized by neck dislocation at different time points (at days 1, 7, 14 and 28).…”

“…According to previous reports, MSCs were isolated from the inguinal fats of juvenile rats (15). Speci cally, after the rats were killed by cervical dislocation, groin fat tissues were sterily separated, cut into 1-mmthick pieces and placed in digestive juices containing 0.05% pancreatic enzymes and 0.1% type I collagen enzyme.…”

“…The identi cation of MSCs was based on previous reports. The phenotypes of the cultured MSCs were identi ed by ow cytometry, and the adiponectic and chondrogenic differentiation abilities of the stem cells were identi ed by oil red O staining and alizarin red staining respectively (15). The freshly-cultured 4th generation MSCs were used in subsequent experiments.…”
