Ingredient selection for plastic composite supports for L-(+)-lactic acid biofilm fermentation by Lactobacillus casei subsp. rhamnosus

Abstract: Plastic composite supports containing 50% agricultural products (oat hulls, soybean hulls, yeast extract, soybean flour, dried bovine erythrocytes, bovine albumin, and/or mineral salts) and 50% (wt/wt) polypropylene were produced by high-temperature twin-screw extrusion. The research employed two half sets of a fivefactorial fractional design (2 5 ؊ 1) to evaluate the effects of different agricultural components on the properties of the plastic composite supports and to select the best plastic composite suppor… Show more

“…At the same time, they are called as microbial cell layers, which are attached on the support materials. Due to high biomass concentration, stability, and potential of long-term fermentation of cells attached on the surface materials, biofilm reactors can be used for production of value-added products such as human lysozyme (Ercan and Demirci 2015b), ethanol (Germec et al 2015(Germec et al , 2016, lactic acid (Ho et al 1997), kojic acid (Liu et al 2016b), isoflavone aglycones (Liu et al 2016a), etc. Nonetheless, microparticles used as a novel approach to improve morphology engineering of filamentous microorganisms and increase their productivity are also applied.…”

“…Repeated-batch fermentations were performed in a bench stirred tank reactor (Sartorius Biostad B Plus, Goettingen, Germany) with a 5-L vessel (working volume of 3-L) with the selected plastic composite support (PCS) material, which consists of polypropylene (50%, w/w), soybean hull (35%, w/w), soybean flour (5%, w/w), yeast extract (5%, w/w), bovine albumin (5%, w/w), and mineral salts at trace quantity [32]. The prepared inoculum (24-h grown culture of S. cerevisiae) was used to inoculate the reactor at 1% (v/v) ratio [5].…”

“…PCS disks employed were a blend of 50% (w/w) polypropylene (Quantum USI Division, Cincinnati, OH), 35% (w/ w) ground soybean hulls (20 mesh) and vacuum-dried (48 h at 110°C, 30 in. of mercury) soybean hulls (Cargill Soy Processing Plant, Iowa Falls, IA), 5% (w/w) yeast extract (Ardamine Z, Universal Flavors), 5% (w/w) defatted soybean flour (Archer Daniels Midland, Decatur, IL), 5% (w/ w) bovine albumin (American Protein Corp., Ames, IA), and mineral salts (2 g of sodium acetate, 1.2 g of MgSO 4 и 7H 2 O, and 0.06 g of MnSO 4 и 7H 2 O per kilogram of mixture) (Ho et al, 1997b). PCS was prepared by high-temperature extrusion in a Brabender PL2000 corotating twin-screw extruder (Model CTSE-V, C. W. Brabender Instruments, Inc., South Hackensack, NJ) through a tubing die assembly (3.2-mm i.d.…”
