Inherited chromosomally integrated human herpesvirus 6 as a predisposing risk factor for the development of angina pectoris

Abstract: Inherited chromosomally integrated human herpesvirus-6 (iciHHV-6) results in the germ-line transmission of the HHV-6 genome. Every somatic cell of iciHHV-6+ individuals contains the HHV-6 genome integrated in the telomere of chromosomes. Whether having iciHHV-6 predisposes humans to diseases remains undefined. DNA from 19,597 participants between 40 and 69 years of age were analyzed by quantitative PCR (qPCR) for the presence of iciHHV-6. Telomere lengths were determined by qPCR. Medical records, hematological… Show more

“…Four microliters (approximately 50 ng) of the DNA was mixed with primers and probes in a final volume of 10 l, mixed with an equal volume of 2ϫ Rotor-Gene multiplex PCR kit (Qiagen), and subjected to 40 cycles of amplification. The conditions for detection of HHV-6A/B were described previously (6,35). Primers and probe used are listed in Table 4.…”

“…At that point in time, cells were detached using trypsin and DNA was isolated and tested by ddPCR for the presence of HHV-6 and the cellular reference gene RPP30, as described previously (82). U65-U66-specific primers were validated previously (12,83) and used for the detection of the integrated HHV-6 genome.…”

“…This condition is termed inherited chromosomally integrated HHV-6 (iciHHV-6) and is present in about 0.2% to 1% of the human population across the world (reviewed in references 10 and 11). In 2015, Gravel et al analyzed DNA samples from 20,000 Quebecers (Canada) for iciHHV-6 and concluded that iciHHV-6A/B represents a predisposing risk factor for angina pectoris (12).…”

“…The effect of iciHHV-6 on human disease is largely unknown, given the challenge of studying relatively rare conditions. One large registry study of 19 597 adults found that angina pectoris occurred 3 times more frequently among individuals with iciHHV-6, 10 perhaps through telomeric disruption or viral gene expression stimulating chronic inflammation.…”
