Inhibiting IL-17A and IL-17F in Rheumatic Disease: Therapeutics Help to Elucidate Disease Mechanisms

Abstract: Purpose of Review Psoriatic arthritis and ankylosing spondylitis belong to a family of rheumatological diseases that lead to painful joint inflammation that impacts on patient function and quality of life. Recent studies have shown that the pro-inflammatory cytokine IL-17 is involved in the inflammatory joint changes in spondyloarthritides. We will review the pathophysiology of IL-17 and review the biological therapies targeting IL-17. Recent Findings IL-1… Show more

“…To this end, expression of both IL-17A as well as IL-17F has been shown to be increased in synovial tissue and entheses of patients with psoriatic arthritis. While data from head-to-head comparator trials are not yet available, indirect evidence suggests comparable efficacies of selective IL-17A inhibition and blockade of IL-17A along with -F ( 43 ).…”

“…While many members of the IL-17 family contribute to inflammation and participate in immune-related diseases and defense against pathogens, they also have various functions in preserving mucosal integrity, responding to allergens, supporting anti-inflammatory responses, and regulating the function of lymphocytes [57]. Among these members, IL-17A and IL-17F are particularly significant in PsA.…”
