Inhibition by Nicotinamide of an Homologous PCA Reaction and Antigen-Induced Histamine Release from Rat Peritoneal Mast Cells

Abstract: The antigen-induced release of histamine both in vitro from rat peritoneal cell suspensions containing mast cells actively sensitized with IgE antibody and in vivo from passively sensitized skin can be inhibited by relatively high doses of nicotinamide. The action of nicotinamide on the cellular mechanism involved in the regulation of antigen-induced histamine release is discussed.

“…Niacinamide has been shown to be a potent inhibitor of cAMP phosphodiesterase and to stabilize mast cells and leukocytes, thereby exerting considerable proteaseand histamine-release blocking activity (27,28). As mentioned previously, histamine and proteases are supposed to take part in the pathogenesis of psoriasis.…”

“…An ability of nicotinamide to give a dose-response inhibition of an in vitro histamine release from sensitized mouse peritoneal mast cells during the reaction with an antigen ( fig. 1) is comparable with its effect in preventing the in vitro antigen-in duced histamine release from isolated rat peritoneal mast cells [31].…”

“…Also, an in vivo protective action of nicotinamide in guinea pig systemic anaphylaxis was observed [2]. Our previous studies have demonstrated an inhibitory activity of nicotinamide on histamine release in IgE-mediated rat anaphy laxis both in vitro (on actively sensitized isolated peritoneal mast cells) and in vivo (on passively sensitized skin) [31]. It has been found that mouse also produces reaginic antibodies [12,16,18,21,23] and recently they have been identified as IgE class of immunoglobulins [17], similar to IgE identified in man [4,5], monkey [7], rabbit [6] and rat [26].…”

“…Thus, it is quite possible that both an inhibitory effect of nicotinamide on PDE and on the decrease of oxidative metabolism in mast cells are responsible for its inhibitory effect on antigen-induced histamine and IgE release [25].…”
