Inhibition of Bcl-2 and Bcl-X enhances chemotherapy sensitivity in hepatoblastoma cells

Abstract: The observed anti-apoptotic phenotype in HB cell lines may contribute to resistance to cytotoxic drugs used in the standard treatment protocol of HB. These pre-clinical results suggest that apoptosis sensitizers with BH-3 mimicry, such as ABT-737, should be further evaluated in preclinical models of HB.

“…However, it remains unclear whether autophagy is beneficial or detrimental for tumor growth. Autophagy serves to eliminate superfluous or damaged organelles and initiates the formation of multiple double-membrane vacuoles that fuse with lysosomes to form intracellular bodies termed ‘autophagosomes’ (28,29). Certain external factors such as nutrient deprivation and specific medications can induce autophagy and lead to decreased cell viability.…”

“…ABT-737 has shown promise in inducing drug sensitivity to multiple chemotherapy drugs in vitro and in animal studies (7, 12, 24). ABT-263, which has a similar specificity profile and efficacy as ABT-737, induces complete tumor regressions in xenograft models of small cell lung cancer and acute lymphoblastic leukemia (13).…”

“…HB cells express high amounts of anti-apoptotic molecules encoded by genes of the Bcl family [11,12]. Bcl-2, an important member of this family, blocks cytochrome C release by sequestering pro-apoptotic BH3-only proteins such as tBid, Bad, Bax, and Bim.…”
