Inhibition of Candidacidal Activity of Polymorphonuclear Cells by Alveolar Macrophage-derived Factor from Lung Cancer Patients

Abstract: Culture supernatants of alveolar macrophages (AM) from lung cancer patients are able to inhibit the candidacidal activity of polymorphonuclear cells (PMN) in vitro. This phenomenon is ascribed to a factor secreted in the culture medium by unstimulated AM from tumor-bearing patients, but not from normal subjects. The inhibitor does not apparently affect the phagocytic activity of PMN, but the superoxide release during phagocytosis is significantly impaired when cells are pretreated with supernatants containing … Show more

“…To prepare peripheral blood monocytes (PBM), heparinized venous blood obtained from normal volunteers was diluted with RPMI 1640. Mononuclear cells were isolated by Ficoll-Hypaque density gradient centrifugation (35). The cells were washed twice in RPMI, placed into cell culture petri dishes (Nunc Inter Med, Roskilde, Denmark) at a concentration of 2 ϫ 10 6 to 3 ϫ 10 6 /ml in RPMI 1640 medium supplemented with 5% FCS (cRPMI) and incubated at 37ЊC in 5% CO 2 for 1 h. The nonadherent cells were removed by washing the dishes three to five times with warm RPMI 1640 medium.…”

“…Serum from patients with solid tumors reduces the cytostatic activity of normal PMN 34. In patients with lung cancer, a factor produced by the alveolar macrophages inhibits the candidacidal activity of PMN 380. A factor that inhibits PMN functions (phagocytosis and the production of ROS) has been detected in plasma from tumor‐bearing rats 381.…”

“…Heparinized venous blood, obtained from healthy donors, was diluted with RPMI 1640. Then, mononuclear cells were separated by Ficoll-Hypaque density gradient centrifugation (32). The cells at the interface were washed twice in RPMI 1640, placed into cell culture petri dishes (Nunc Inter Med, Roskilde, Denmark) at a concentration of 2 ϫ 10 6 to 3 ϫ 10 6 /ml in RPMI 1640 supplemented with 5% fetal calf serum, and incubated at 37ЊC in 5% CO 2 for 1 h. The nonadherent cells were removed by washing the dishes three to five times with warm RPMI 1640.…”
