Inhibition of interaction between Staphylococcus aureus α-hemolysin and erythrocytes membrane by hydrolysable tannins: structure-related activity study

Abstract: The objective of the study was a comparative analysis of the antihemolytic activity against two Staphylococcus aureus strains (8325-4 and NCTC 5655) as well as α-hemolysin and of the membrane modifying action of four hydrolysable tannins with different molecular mass and flexibility: 3,6-bis-O-di-O-galloyl-1,2,4-triO -galloyl-β-d-glucose (T1), 1,2,3,4,5-penta-O-galloyl-β-d-glucose (T2), 3-O-galloyl-1,2-valoneoyl-β-d-glucose (T3) and 1,2-di-O-galloyl-4,6-valoneoyl-β-d-glucose (T4). We showed that all the compou… Show more

“…Interestingly, the berry polyphenolic fraction completely inhibited the hemolytic activity of P. gingivalis and as such, may impede its ability to proliferate. Although the mechanism involved in the inhibition of the P. gingivalis hemolytic activity by the berry polyphenolic fraction has not been investigated, it may be related to structural changes in erythrocyte membranes caused by the polyphenols thus limiting the binding of the hemolysin, as previously reported in Staphylococcus aureus [ 32 ].…”

“…We measured the fluorescence anisotropy values for TMA-DPH probe locating in the outer area of the membrane at the level of the fourth carbon atom and for DPH probe inserting itself in the membrane hydrophobic region in the middle of the lipid bilayer of red blood cells. The fluorescence anisotropy values were used to calculate the lipid order parameter [14,16,17] (S) and were presented as a relative change (S/S 0 ) in the presence of compounds compared to control (Figure 4C, D). These results reveal increased surface lipid ordering based on higher TMA-DPH S/S 0 values and lower lipid ordering in the middle of bilayer due to lower DPH S/S 0 values in the presence of both molecules.…”

“…Strong antioxidant and radical scavenging capacities make HTs play a role in the treatment of various diseases. The antioxidant activity and scavengers of hydroxyl, superoxide, and peroxyl radicals largely depend on their structure [ 2 , 3 , 4 ]; for example, an increase in anti-radical effects was observed with an increase in the degree of polymerization [ 5 ]. Modern medical research has shown that HTs have pharmacological activities against COVID-19 [ 6 ], bacteria [ 4 ], inflammation [ 7 ], nephropathy [ 8 ], diabetes [ 9 ], HIV, and many other pathologies [ 5 , 10 , 11 ].…”
