Inhibition of Intercellular Adhesion Molecule-1 Protein Expression by Antisense Oligonucleotides Is Neuroprotective After Transient Middle Cerebral Artery Occlusion in Rat

Abstract: Background and Purpose-The present study was performed to determine whether antisense inhibition of intercellular adhesion molecule-1 (ICAM-1) protein expression decreases focal ischemic brain damage. Methods-Male spontaneously hypertensive rats underwent 1-hour middle cerebral artery occlusion (MCAO) and 24-hour reperfusion.

“…Total RNA (1 mg) from each sample was reverse transcribed with oligo(dT) 15 and random hexamer primers using M-MuLV reverse transcriptase (Life Technologies, Rockville, MD, USA). Real-time PCR was conducted as described earlier (Vemuganti et al, 2004). Ten nanograms of cDNA and MCP-1 primers (5 0 -3 0 , CCA GAA ACC AGC CAA CTC TCA and GGC AGC AAC TGT GAA CAA CAG) were added to SYBR Green PCR Master Mix (SYBR Green I Dye, AmpliTaq s DNA polymerase, dNTPs with dUTP and optimal buffer components; Applied Biosystems, Foster City, CA, USA) and subjected to PCR amplification (one cycle at 501C for 2 min, one cycle at 951C for 10 min, and 40 cycles at 951C for 15 s and 601C for 1 min) in a TaqMan 5700 Sequence Detection System (Applied Biosystems).…”

“…MCP-1 (500 ng/ml) or aCSF with 0.1% BSA was infused into the striatum (coordinates: anteriorposterior À0.3 mm, lateral 2.5 mm, dorsal-ventral 4 mm) of 6 normal rats using Alzet osmotic minipumps (Model 2001, DURECT Co.) with a flow rate of 1 mL/h for 7 days as described by our previous studies (Dempsey et al, 2003;Vemuganti et al, 2004). Rats were killed at day 7 of infusion.…”

“…Quantitative real-time PCR was conducted on the same RNA samples used for the GeneChip analysis as described previously (Vemuganti et al, 2004). Total RNA (2 mg) from each sample was reverse-transcribed with oligo(dT)15 and random hexamer primers using M-MuLV reverse transcriptase (Life Technologies).…”
