Inhibition of Lipid Peroxidation in Synaptosomes and Liposomes by Nitrates and Nitrites

Abstract: NO is produced endogenously from arginine by the action of NO synthase, and exogenously by nitrovasodilators, including organic nitrates and nitrites. NO has been proposed as a cytotoxic and cytoprotective agent. There is strong evidence that NO acts as an apparent antioxidant in inhibiting lipid peroxidation, via chain termination, and interestingly lipid nitrates and nitrites have been proposed to be products of this chain termination. Both pro- and antioxidant mechanisms may be drawn for nitrates and nitrit… Show more

“…Like in our case, in the majority of experiments ELF-MF increases lipid peroxidation [16], [53], [60], [65]. We can propose that ELF-MF, through increasing the level of NO, is involved in the reduction of ILP in ischemic gerbils, because NO itself may directly inhibit lipid peroxidation by intercepting alkoxyl and peroxyl radical intermediates and thus terminating chain propagation reaction [66], [67]. Di Loreto et al [18] also proposed that ELF-MF can simultaneously activate pro- and antioxidants.…”

“…Disulfides, notably diallyl disulfide (DADS), have been studied as garlic-derived chemopreventive agents that reduce ACF formation in the rat/azoxymethane model (23,24). Reports suggest that the disulfide group of DADS is responsible for the antiproliferative activity (16). DADS was reported to induce a G 2 -M phase cell cycle block in HCT-15 cells (25) and to be anti-antiproliferative in Caco-2 cells (24, 26).…”

“…Reaction of nitric oxide with superoxide causes formation of peroxynitrite that initiates lipid peroxidation via reaction of lipids with its decomposition products hydroxyl radical and nitrogen dioxide (Brookes et al, 1998;Rubbo et al, 1994). In contrast, nitric oxide itself may directly inhibit lipid peroxidation by intercepting alkoxyl and peroxyl radical intermediates thereby terminating chain propagation reactions (Nicolescu et al, 2002;Niziolek et al, 2003;Rubbo et al, 1994).…”
