Inhibition of neuronal FLT3 receptor tyrosine kinase alleviates peripheral neuropathic pain in mice

Abstract: Peripheral neuropathic pain (PNP) is a debilitating and intractable chronic disease, for which sensitization of somatosensory neurons present in dorsal root ganglia that project to the dorsal spinal cord is a key physiopathological process. Here, we show that hematopoietic cells present at the nerve injury site express the cytokine FL, the ligand of fms-like tyrosine kinase 3 receptor (FLT3). FLT3 activation by intra-sciatic nerve injection of FL is sufficient to produce pain hypersensitivity, activate PNP-ass… Show more

“…PDGF peptides can bind with extremely high affinity to the VEGFR-2, and computational predictions suggest that PDGF peptides may account for a large percentage of VEGFR-2 binding in vivo (Mamer et al, 2017). Also, the Flt3 RTK can alter mechanical sensitivity in a TRP channel-dependent manner (Rivat et al, 2018), further supporting the concept that RTKs may regulate mechanical nociception. Further studies are indicated to better understand how RTKs, piezo, and TRP channels might interact to regulate mechanical sensitivity in vertebrates.…”

“…Therefore, the test can be utilized to elucidate the potential mechanism of tested drugs. CNS acting compounds act on both phases; however, many anti-inflammatory drugs inhibit only the chronic phase [24,25].…”

“…The sections were washed 3 times, slidemounted, and coverslipped in mounting medium with the nuclear stain DAPI (Vector Laboratories). Anti-Tuj1 antibody has previously been used for the detection of DRG neurons (Rivat et al, 2018). Anti-Tuj1 antibody reacts with a structural protein that is a component of tubulin, whereas anti-NeuN antibody stains the nuclei of neurons.…”
