Inhibition of Retinal Neovascularization by Intravitreal Injection of Human rPAI-1 in a Rat Model of Retinopathy of Prematurity

Abstract: PAI-1 inhibits pathologic angiogenesis without adversely affecting normal vasculogenesis, an attractive feature for ROP therapies. Moreover, PAI's relationship to matrix metalloproteinases, which are also implicated in angiogenesis, suggests that the proteolytic aspect of the process may provide additional downstream therapeutic targets.

“…The vitreous volume of the 21-day-old rats in our experiment is 27.8 8 0.6 l. This is also close to the reported vitreous measurement, which is 25 l for the 18-day-old rats [40] . These values suggest that our results are reliable and can be used as a reference for eye research.…”

“…The vitreous volume in adult rats has been widely reported, but the measurements varied widely, from 13, 40, 56, 60, 100 to 120 l in different papers [8,[20][21][22][34][35][36][37][38] . In younger rats, the vitreous volume was reported as 6.8 l at age 21 days [8] , 30 and 25 l at 18 days [39,40] , 20 and 11 l at 14 days [41,42] and 18 l at 12 days [39] . Although intravitreal injections have been made in younger rats, from days 1 to 9, no data can be found in the literature on vitreous volume for rat pups younger than 12 days [43] .…”

“…Multiple gene products we find secreted by the liver have been implicated in ameliorating blood vessel destruction, such as EPO, PAI-1, orosomucosoid, adrenomedulin, apelin, VEGF, and angiopoietin-like protein 3, implying that the power of targeting a transcription factor lies in the coordinated up-regulation of multiple proteins necessary for organized blood vessel growth (31,(36)(37)(38)(39)(40)(41). We have also considered that the differences in retinal RNA induced by DMOG or Roxadustat demonstrate HIF-1α or HIF-2α dependence.…”

“…It causes less effusion than cryotherapy and has at least as good results from an anatomical and visual standpoint (10)(11)(12) . In the not so distant future, it may be that pharmacologic therapy will be possible (7,(13)(14)(15)(16) . Besides the immediate short-term results it seems that laser therapy may also be beneficial in respect to longterm functional and visual outcome.…”
