A new pathway of dimethylsulfide (DMS) metabolism was identified in a novel species of Gammaproteobacteria, Methylophaga thiooxidans sp. nov., in which tetrathionate (S(4)O(6)(2-)) was the end-product of DMS oxidation. Inhibitor evidence indicated that DMS degradation was initiated by demethylation, catalysed by a corrinoid demethylase. Thiosulfate was an intermediate, which was oxidized to tetrathionate by a cytochrome-linked thiosulfate dehydrogenase. Thiosulfate oxidation was coupled to ATP synthesis, and M. thiooxidans could also use exogenous thiosulfate as an energy source during chemolithoheterotrophic growth on DMS or methanol. Cultures grown on a variety of substrates oxidized thiosulfate, indicating that thiosulfate oxidation was constitutive. The observations have relevance to interactions among sulfur-metabolizing bacteria in the marine environment. The production of tetrathionate from an organosulfur precursor is previously undocumented and represents a potential step in the biogeochemical sulfur cycle, providing a 'shunt' across the cycle.

Sodium tetrathionate (TT) was assessed for its possible protection of proteolytic activity (PA) of rehydrated crude papain during drying and storage. The addition of 1% 'IT significantly decreased the loss of PA of rehvdrated crude papain after being oven dried at 55°C. This protection of PA was I.?-20% higher than that from metabisulfite. Crude oauain with 1% 'PI lost 20% of PA and with metabisulfite lost 45% L I of PA in 13 wk at room temperature. Results suggest 'IT may decrease loss of PA during papaya latex processing and storage of crude papain.

The metabolism of C 1 organosulfur compounds by the Bacteria is important in the biogeochemical cycling of sulfur and carbon and in climate regulation in terms of mediating release of, e.g., dimethylsulfide from the oceans. Herein we review the canon of work on the metabolism of dimethylsulfide, dimethylsulfoxide, dimethylsulfone, methanesulfonate, dimethyldisulfide, and methanethiol, in terms of dissimilation to formaldehyde or carbon dioxide when used as carbon and energy sources by methylotrophs or autotrophs, oxidation to sulfite prior to assimilation as sulfur sources, and use as respiratory terminal electron acceptors. We discuss the enzymology of the metabolism of these compounds and propose a revision to the Enzyme Commission classification to some of them where multiple enzymes are clearly grouped under one name at present. We also provide methodologies for enzyme assays, for the safe handling and quantification of these compounds, and for the synthesis of carbon-14, carbon-11, sulfur-34, and sulfur-34 compounds for use in physiological and ecological studies.
