Inhibitory Effect of Delphinidin on Oxidative Stress Induced by H2O2 in HepG2 Cells

Abstract: Chronic liver diseases (CLDs) are correlated with oxidative stress induced by the accumulation of intracellular reactive oxygen species (ROS). In this study, we employed HepG2, a human liver carcinoma cell line containing many antioxidant enzymes, to explore the function of delphinidin against oxidative stress induced by H2O2 and to provide scientific data of the molecular mechanism. Cells were pretreated with different concentrations of delphinidin (10 μmol/L, 20 μmol/L, and 40 μmol/L) for 2 h before treatmen… Show more

“…Dp promotes Nrf2 nuclear translocation and leads to increase expression of antioxidant protein HO-1, which is an Nrf2-related phase II enzyme heme oxygenase-I present in HepG2 cells. This confirms the hepatoprotective effect of Dp and its role in regulation of the expression of Nrf2/HO-1 to protect HepG2 cells ( 151 ). Dp has been demonstrated to decrease hepatic inflammatory markers such as TNF-α, IL6, and INF.…”

“…4). 108,118 Delphinidin-treated (10.0, 20.0, and 40.0 µmol L −1 , for 24 hours) HepG2 cells showed a reduction in H 2 O 2 -induced oxidative stress by modulating a Nrf2-dependent mechanism, similar to CGA findings, suggesting that isolated delphinidin might be a reliable strategy against NAFLD (Table 2). 106,118 Cyanidin-3-glucoside is the major cyanidin-derivative compound, which is suggested to play a similar role to delphinidin, modulating the oxidative stress axis in both in vitro and in vivo models.…”

“…Due to their antioxidant potential, delphinidin glucosides showed the strongest scavenging activity against superoxide anion and peroxynitrite among several anthocyanins [ 32 ]. Moreover, delphinidin was reported to exert cytoprotective effects for human chondrocytes against oxidative stress [ 33 ], as well as inhibitory effects on oxidative stress in HepG2 cells [ 34 ]. To the best of our knowledge, this is the first study reporting malvidin-3-O-glucoside in A. unedo wild fruit extracts.…”
