Inhibitory effects of bFGF, VEGF and minoxidil on collagen synthesis by cultured hair dermal papilla cells

Abstract: Dermal papilla cells of rat vibrissa follicles cultivated in monolayers and in three-dimensional collagen gels show a different morphology in these culture systems. Dermal papilla cells cultured in lattices tend to express morphological features resembling those seen in vivo. Quantification of total collagen by incorporation of 3H-proline in monolayer cultures and in collagen lattices show that the amount of collagen found in dermal papilla cells is higher than that secreted. Moreover, collagen synthesis measu… Show more

“…HaCaT cells were seeded into 96-well plates at 1.5ϫ10 4 cells/well, cultured for 24 h in serum-free DMEM, and treated with the vehicle (0.12 mM HCl/DMSO diluted 1 : 1000 in serum-free DMEM) as control, minoxidil (0.01 mM), retinol (1.7 pM-1.7 nM), or minoxidil (0.01-1.0 mM) plus retinol (1.7 pM-1.7 nM) for 5 d. Cell growths and viabilities were measured by adding 0.4% trypan blue in 0.9% saline to a 50% dilution, and cells were counted using a hemocytometer. Western Blot Confluent DPCs and HaCaT were incubated for 24 h treated with vehicle, minoxidil (0.01 mM), retinol (1.7 pM), or 0.01 mM minoxidil plus 1.7 pM retinol for 1 h to evaluate Erk and Akt expression and for 24 h to evaluate Bcl-2/Bax ratios and b-actin expression.…”

“…In addition, VEGF is active on DPC metabolism. It affects collagen 11 and eicosanoid 12 production and thus may be implicated in hair growth through these physiological and metabolic pathways.…”

“…However, lengthier body hairs in individuals using minoxidil would indicate that anagen duration may also be prolonged . There is confusion as to the exact mechanism by which minoxidil works; opening potassium ion channels promoting vasodilation, upregulation of VEGF and upregulation of prostaglandin E2 have all been proposed . Regardless, if minoxidil simply accelerates hair cycle progression and does not impact efflux or influx, it would only slow the progression of miniaturisation and/or reduce the appearance of baldness by increasing the number of hair follicles in anagen at any given point in time.…”
