Inhibitory effects of probiotic Bacillus coagulans against MCF7 breast cancer cells

Abstract: Background and Objectives: Secondary metabolites in the supernatants of probiotic microorganisms have shown antican- cer effects. The present study was aimed to investigate the cytotoxicity of Bacillus coagulans supernatants and their role in apoptosis induction in MCF7 cancer cells. Materials and Methods: The inhibition of MCF7 cancer cells by Bacillus coagulans supernatants was assessed by MTT assay at three exposure times of 24, 48, and 72 h. Apoptosis induction was explored by flow cytometry while th… Show more

“…To finish the synthesis of metabolites, 10% of the overnight culture was inoculated into an MRS broth medium and cultured for 72 h. Centrifugation was carried out at 6000 rpm (Hettich, Germany), at room temperature, for 20 min to obtain the bacterial culture supernatant. Then, 100 mL of the culture medium was thickened with a freeze dryer to about 20 mL and stored in a refrigerator 35 .…”

“…First, SKBR3 cells were prepared in a 6-well plate as in the previous step and treated with IC50 of bacterial culture supernatant for 48 h. Untreated cells were considered negative controls. RNA extraction, cDNA synthesis, and realtime PCR were performed according to the protocol reported in the previous study (27).…”
