Climate changes due to global warming affect the productivity of forage crops such as maize. However, the productivity of maize could be maximized by improving the plant’s adaptation and tolerance through breeding, using tissue culture. This study aims to determine the effect of explant types and light conditions on callus induction of srikandi putih maize. It was conducted at the Laboratory of Forage and Pasture Science, Faculty of Animal Science, Universitas Gadjah Mada, Yogyakarta. Furthermore, it was carried out using a completely randomized design in a factorial pattern. There were two treatment factors namely explant types, which consisted of roots, stems, and leaves, and light conditions consisting of darkness and light. The results showed that callus induction occurred in the stem and root of explants at 60%, but not in the leaves. It also showed that the stem explants produced a higher fresh weight of callus at 239.6 mg compared to the roots which produced 77.6 mg. The dark condition also produced a 49.3% higher amount of callus with fresh weight of 115.1 mg. Meanwhile, the light condition produced 30.7% higher amounts of callus with fresh weight of 96.3 mg. It was concluded that all treatments showed a compact texture of callus, but the use of stem explants in dark conditions was the best treatment.

Tomatoes (Lycopersicon esculentum Mill.) are a source of carotenoids they are easy to find. These compounds function as precursors of vitamin A, antioxidant, and prevent cancer. The extraction of carotenoid compounds for commercial products usually uses fresh plants, which are less efficient and require a lot of raw materials. The supply of these raw materials can be done through tissue culture. The frequency of subculture or supply of nutrients in tissue culture is very influential on the content of callus carotenoids produced. This study aimed to determine the effect of subculture frequency on growth, development and callus carotenoid content and to find out the right frequency of subculture to produce callus with optimal growth, development and carotenoid content. The design this study was a single Completely Randomized Design (CRD) with 4 treatments of subculture frequency and 5 replications. The data obtained were analyzed by ANOVA at the 95% test level followed by DMRT in case a significant different was found The results showed subculture frequency affected growth, development and carotenoid content of callus Lycopersicon esculentum. Mill. The most optimal treatment to induce growth and production of carotenoids in this study was treatment of thrice subculture while the most optimal treatment in inducing development was the twice subculture treatment.

Onion is one of the primary commodities of growing in Central Sulawesi and is a raw material for the fried shallot processing industry cv. lembah palu. This study aimed to determine the effect of seaweed extract and NPK on the growth and yield of the onion. This research was performed at the Faculty of Agriculture farm, Tadulako University. This research was prepared using a Randomized Block Design (RBD) consisting of seven treatments, with the following details: (A1) = Caulerpa sp Extract 100 ml/pot; (A2) = Sargassum sp Extract 100 ml/pot; (A3) = Ulva sp. Extract 100 ml/pot; (A4) = 0.25 g NPK fertilizer per plant; (A5) = 100 ml of Caulerpa sp. Seaweed Extract and 0.25 g NPK per pot; (A6) = Sargassum Sp Extract 100 ml and 0.25 g NPK per pot; (A7) = Ulva sp. Extract 100 ml and 0.25 g NPK per pot. Thus, there are seven treatments. Each treatment is repeated three times so that there are 21 experimental units. The results showed that the treatment of seaweed extract and NPK obtained better number of leaves, plant height, fresh plant weight, fresh tuber weight, plant dry weight, and number of tuber clumps.
