A substitution of the polyhedrin of Autographa californica nucleopolyhedrovirus (AcNPV) with that of Spodoptera exigua NPV (SeNPV) and Bombyx mori NPV, respectively, resulted in morphological changes of the polyhedra. Morphological features and quantity of polyhedra of recombinant AcNPVs were examined and compared in three cell lines: Spodoptera frugiperda (Sf9), S. exigua (Se301) and Trichoplusia ni (Hi5) cells. The shape of the polyhedra produced by each virus did not change significantly according to the cell line. The size of the polyhedra differed, however, with the largest in Se301 cells, followed by Hi5 and Sf9 cells in decreasing order. The yield of polyhedra and the appearance of polyhedra-positive cells in each cell line also showed significant differences. Especially notable was the change of polyhedra shapes from irregular to regular in the recombinant AcNPV containing SeNPV polyhedrin in all cell lines. These results suggest that the shape and uniformity of polyhedra depend on some viral gene other than the polyhedrin gene. It is also suggested that there is a host cell factor that is responsible for the determination of polyhedra size.

The nucleotide sequence of a 1.1 kbp fragment of the multiple nucleocapsid nuclear polyhedrosis virus (MNPV) of Spodoptera exigua (Se) containing the polyhedrin gene was determined. An open reading frame (ORF) of 738 nucleotides (nt) was detected. This ORF encoded a protein of 246 amino acids with a predicted Mr of 29K. The nucleotide and amino acid sequences were compared with the sequences of eight other NPV polyhedrins. The SeMNPV polyhedrin protein was most closely related to S. frugiperda MNPV polyhedrin with differences in only five amino acids, and most distantly related to the Lymantria dispar MNPV polyhedrin. The size of the mRNA was approximately 1000 nt, as determined by Northern blot analysis. Using primer extension assays and S1 nuclease mapping the transcriptional start and stop sites of the polyhedrin mRNA were located. The 5' regulatory sequence appeared to be 44 nt in length with the mRNA start site predominantly at the first A of the TAAG consensus start sequence. Two degenerate poly(A) signals were found immediately downstream of the translational stop signal. The transcriptional stop was located approximately 230 nt downstream from the translational stop signal, in an AT-rich sequence that appears to be common to all baculovirus polyhedrin genes. The SeMNPV polyhedrin mRNA does not appear to be polyadenylated.

The Rachiplusia ou multiple nucleopolyhedrovirus (RoMNPV) is a variant of Autographa californica MNPV (AcMNPV) but is significantly more virulent against several major agricultural pests. The genome sequence of the R1 strain of RoMNPV was determined and compared to that of AcMNPV strain C6. The RoMNPV genome is approximately 131?5 kbp with a G+C content of 39?1 %. The homologous repeat regions (hrs) described for AcMNPV-C6 are present in RoMNPV-R1 but the hrs of RoMNPV have fewer palindromic repeats. The RoMNPV-R1 nucleotide sequence is almost completely collinear with the sequence of AcMNPV-C6 and contains homologues of 150 of the 155 ORFs described for AcMNPV-C6. Deletions, insertions and substitutions have resulted in the loss of homologues for AcMNPV ORFs ac2 (bro), ac3 (ctl), ac97, ac121 and ac140 from the RoMNPV genome. The average amino acid sequence identity between RoMNPV and AcMNPV ORFs is 96?1 % and there are differences in promoter motif composition for 23 of these ORFs. Maximum-likelihood analysis of selection pressures on AcMNPV and RoMNPV ORFs indicate that ORFs ro18/ac20-ac21 (arif-1) and ro135/ac143 (odv-e18) have undergone positive selection.
