Insights into the Cardiotoxic Effects of Veratrum Lobelianum Alkaloids: Pilot Study

Abstract: Jervine, protoveratrine A (proA), and protoveratrine B (proB) are Veratrum alkaloids that are presented in some remedies obtained from Veratrum lobelianum, such as Veratrum aqua. This paper reports on a single-center pilot cardiotoxic mechanism study of jervine, proA, and proB in case series. The molecular aspects were studied via molecular dynamic simulation, molecular docking with cardiac sodium channel NaV1.5, and machine learning-based structure–activity relationship modeling. HPLC-MS/MS method in combinat… Show more

“…Veratrum lobelianum is a robust herbaceous plant found in Northeast Asia, Central Europe, and North America that is known for its toxicity in humans and animals [33]. The alkaloid jervine, abundant in V. lobelianum, exhibits antitumor effects and influences DNA damage repair in nasopharyngeal carcinoma cells [24,27,34]. Our experiments, using jervine-containing V. lobelianum extract, activated the DNA damage checkpoint by downregulating hedgehog signaling, suggesting a complex relationship (Figures 3 and 4).…”

“…We hypothesized that the compounds present in the herbs may contribute to the observed phenotypes in the herbal extracts. Jervine, a primary alkaloid found in V. lobelianum [ 23 , 24 ], has been identified as an inhibitor of the Hedgehog pathway in nasopharyngeal carcinoma—a crucial cellular pathway involved in cell growth, differentiation, and tissue formation [ 25 , 26 , 27 ].…”

“…According to the literature, gervin has anti-tumor [41,42], anti-inflammatory, analgesic [43], and radioprotective [44] activities. There is a study investigating the cardiotoxic mechanism of gervin, protoveratrin A, and B [45].…”
