Intact glycosphingolipidomic analysis of the cell membrane during differentiation yields extensive glycan and lipid changes

Abstract: Glycosphingolipids (GSLs) are found in cellular membranes of most organisms and play important roles in cell-cell recognition, signaling, growth, and adhesion, among others. A method based on nanoflow high performance liquid chromatography-chip-quadrupole-time-of-flight mass spectrometry (nanoHPLC Chip-Q-TOF MS) was applied towards identifying and quantifying intact GSLs from a variety of samples, including cultured cell lines and animal tissue. The method provides the composition and sequence of the glycan, a… Show more

“…Furthermore, identified gangliosides by LC-MS/MS were reconfirmed using retention time prediction, as described in the literature [28,34]. For example, Figure 1c showed very high relationship ( R 2 = 0.9993) among GM1 compounds with one double bond in NCI-H358.…”

“…Therefore, to address the complexity of cell surface gangliosides, development of novel and rapid analytical platform is required. Highly sensitive LC/MS technology has recently emerged as an effective tool for characterizing large numbers of intact gangliosides [28,29,30]. However, full characterization of structural complexity of gangliosides is still remained to be addressed.…”

“…Assessment of Laurdan emission in these studies showed a decrease of the GP value with the increased skin depth, from stratum corneum through to the dermis, including a progressive decrease at the level of the epidermis; this is in line with data presented here. While we utilized Laurdan to assess differentiation process in epidermal keratinocytes specifically, we anticipate that this methodology may be also potentially successfully used with other cell types, which undergo lipid remodeling during differentiation or maturation (Reynier et al, 1991;Wong et al, 2018).…”

“…Recently, Wong et al [143] developed a method for extensive identification and quantitation of cell membrane GSLs with nano HPLC-chip-Q-TOF MS, with which more than 200 intact GSLs of differentiated Caco-2 cell line were characterized (Figure 5). With tandem MS, the composition and connectivity of glycan head groups were profiled, together with their attached ceramides with varying numbers of hydroxyl groups, lipid lengths, and degrees of unsaturation.…”
