Integrated Bayesian species delimitation and morphological diagnostics of chorioptic mange mites (Acariformes: Psoroptidae: Chorioptes)

Abstract: The external morphology of adult and immature stages of mange mites of the genus Chorioptes was investigated with the aid of light and scanning electron microscopy. A molecular phylogeny of this genus was inferred based on six genes (18S, 28S rDNA, EF1-α, SRP54, HSP70, and CO1). The validity of four species (Ch. bovis, Ch. panda, Ch. texanus, and Ch. sweatmani sp. nov. described from the moose from Sweden, Finland, and Russia) was confirmed based on morphology and a Bayesian species delimitation analysis incor… Show more

“…For 173 taxa we sequenced six genes, 18S ribosomal RNA gene (18S), 28S ribosomal RNA gene (28S), elongation factor 1alpha100E Ef1alpha100E (EF1‐α), signal recognition particle protein 54k Srp54k (SRP54), heat shock protein cognate 5 Hsc70‐5 (HSP70), cytochrome c oxidase subunit I (COX1), using previously published amplification, sequencing, and DNA extraction protocols (Klimov and OConnor ; Knowles and Klimov ; Klimov and OConnor ; Bochkov et al. ). Our aligned matrix had 11,468 sites and did not have missing data due to amplification/sequencing failures.…”

“…This query yielded 284 results, from which we only selected studies using BPP on arthropods (20 total). We did not include a study on parasitic mites (Bochkov et al 2014) because estimates of host-specific mites are included separately in our analyses of arthropod richness (Appendix S2).…”

“…Six genes (18S, 28S rDNA, EF1-a, SRP54, HSP70, and CO1) were sequenced. DNA extraction, rDNA secondary structure alignment, oligonucleotide primers, amplification and sequencing were as described previously (Klimov and OConnor, 2008;Knowles and Klimov, 2011;Bochkov et al, 2014b). Outgroups were chosen based on our previous, large-scale molecular study designed to elucidate the origin of house dust mites (Klimov and OConnor, 2013).…”
