Integrated multi-omics analysis identifies CD73 as a prognostic biomarker and immunotherapy response predictor in head and neck squamous cell carcinoma

Abstract: BackgroundAdvances in tumor immunotherapy have been developed for patients with advanced recurrent or metastatic (R/M) HNSCC. However, the response of most HNSCC patients to immune checkpoint inhibitors (ICI) remains unsatisfactory. CD73 is a promising target for tumor immunotherapy, but its role in HNSCC remains insufficient. In this study, we aim to explore the function of CD73 in HNSCC.MethodsTranscriptomic and clinical data of TCGA-HNSC were downloaded from UCSC Xena for analysis of CD73 mRNA expression an… Show more

“…As shown in Fig. 6, immune functions such as APC_co_stimulation, CCR, Parain ammation, and MHC_class_I were signi cantly up-regulated in high-risk group, while Type_II_IFN_Reponse and Cytolytic_activity were signi cantly down regulated.TIDE, as a new computing architecture, represents tumor immune dysfunction and rejection, and is used to evaluate the possibility of tumor immune escape [19][20]. As illustrated in Fig.…”

“…Tumor Immune Single-cell (TISCH) database is an available resource facilitates the exploration of the tumor microenvironment (TME) via single-cell RNA sequencing (scRNA-seq) data ( 13 , 14 ). We analyzed the distribution and expression of TBC1D1 in diverse cell types within the glioma microenvironment using the TISCH.…”

“…Tissue samples from patients with LUAD were scored using Cell-type Identification by Estimating Relative Subsets of RNA Transcripts (CIBERSORT), microenvironment cell populations (MCP)-counter, and estimation methods ( 14 , 15 ). Pearson correlation analysis was used to explore the relationship between the immune score and the levels of tumor purity, immune cells, and immune cell markers, and P<0.05 was considered a significant screening criterion.…”
