Integrating environmental DNA monitoring to inform eel (Anguilla anguilla) status in freshwaters at their easternmost range—A case study in Cyprus

Abstract: Despite significant population declines and targeted European Union regulations aimed at Anguilla anguilla conservation, little attention has been given to their status at their easternmost range. This study applies wide‐scale integrated monitoring to uncover the present‐day eel distribution in Cyprus' inland freshwaters. These are subject to increasing pressures from water supply requirements and dam construction, as seen throughout the Mediterranean. We applied environmental DNA metabarcoding of water sample… Show more

“…Library preparation was carried out following the methodology outlined in Griffiths et al (2023), summarised below: Nested metabarcoding following a two-step PCR protocol was performed, using multiplex identification (MID) tags in both steps to enable sample identification as described in (Kitson et al, 2019). The first PCR (PCR1) was performed in triplicate (3× PCR replicates per eDNA extract), amplifying a 106bp fragment using published 12S ribosomal RNA primers 12S-V5-F (5′-ACTGGGATTAGATACCCC-3′) and 12S-V5-R (5′-TAGAACAGGCTCCTCTAG-3′) (Kelly et al, 2014;Riaz et al, 2011).…”
