Integration of cytogenetic and genetic linkage maps of Lotus japonicus, a model plant for legumes

Abstract: To construct a high-resolution pachytene chromosome map, we used the chromosome image analyzing system version 3 and fluorescence in situ hybridization. Two ribosomal RNA genes (45S rDNA and 5S rDNA), two major tandem repeat DNAs (LjTR1 and LjTR2), two major retroelements (LjRE1 and LjRE2), and 27 transformation-competent artificial chromosome clones were physically localized on Lotus japonicus (Miyakojima MG-20, 2n = 12) chromosomes. The distributions of heterochromatin and euchromatin along six chromosomes w… Show more

“…With the construction of a genetic linkage map in Passiflora (Carneiro et al, 2002;Oliveira et al, 2008), genetic sequences of agronomic importance could be located on the chromosomes by the FISH technique, resulting in the integration of the cytogenetic and genetic linkage map, as noted in Lotus japonicus (Ohmido et al, 2010). With the utilization of Passiflora interspecific hybrids in the ornamental plant market (Abreu et al, 2009), the use of the GISH technique, still unpublished for this genus, is promising with regard to the identification of introgression of chromosomes or chromosomal fragments.…”

“…The 5S rDNA and telomere repeat sequences were labeled by PCR with biotin-16-dUTP or digoxigenin-11-dUTP (Roche, Branchburg, N.J., USA), and the 16 BAC clones and 26S rDNA sequence were labeled using a Nick Translation kit (Roche) with biotin-16-dUTP or digoxigenin-11-dUTP according to the method described by Ohmido et al [2010].…”

“…FISH analysis using the 2 rRNA genes, the telomeric repeat, and the BAC clones was performed using chromosome spreads carefully prepared according to the method described by Ohmido et al [2010]. For immunocytochemical fluorescence detection, the slides were incubated with fluorescein isothiocyanate (FITC)-conjugated anti-digoxigenin antibody (Roche) and streptavidin-conjugated Cy3 (Cy3-SA; Jackson ImmunoResearch Laboratories, West Grove, Pa., USA) in 4 !…”

“…Lotus japonicus is a perennial species closely related to birdsfoot trefoil (L. corniculatus), an important forage legume. The studies performed in L. japonicus include wholegenome sequencing, molecular-marker development, and the construction of high-density linkage maps (Sato et al 2008;Ohmido et al 2010). Three linkage maps have been reported for the species, including one map based on the intraspecific cross of L. japonicus Gifu  L. japonicus MG20 (Hayashi et al 2001;Sato and Tabata 2006), another based on the interspecific cross of L. filicaulis  L. japonicus Gifu , and a third map under construction from the interspecific cross of L. japonicus Gifu  L. burttii (Kawaguchi et al 2005;Sandal et al 2012).…”
