Integrative metabolomics reveals unique metabolic traits in Guillain-Barré Syndrome and its variants

Abstract: Guillain–Barré syndrome (GBS) is an acute fatal progressive disease caused by autoimmune mechanism mainly affecting peripheral nervous system. Although the syndrome is clinically sub-classified into several variants, specific biomarker and exact pathomechanism of each subtypes are not well elucidated yet. In current study, integrative metabolomic and lipidomic profiles were acquisitioned from cerebrospinal fluid samples of 86 GBS from three variants and 20 disease controls. And the data were systematically com… Show more

“…There are only a few reports in the literature that analysed CSF lipids in MS or GBS. These studies were targeted either on SM analysis in demyelinating disorders of the PNS 26 , on the assessment of the sphingolipidome 27 or the global lipidome with a fatty acid-targeted approach 28 in patients with MS, or on the polar metabolome from patients with MS 29 and GBS 30 .…”

“…Transfer line and ion source temperatures were set to 280°C and 250°C, respectively. Mass spectra were collected ranging from 85 to 500 m/z at a scan rate of 20 spectra/s with a detector voltage of 1850 V. 15 …”

“…The aliquots were concentrated to complete dryness in a speed vacuum concentrator (SCANVAC, Korea). 49 The dried cecal extracts were derivatized with 5 µl of 40mg/ml methoxyamine hydrochloride (Sigma-Aldrich, St. Louis, MO, USA) in pyridine (Thermo, USA) and then incubated for 90 min (800 rpm at 30°C). After the first derivatization step, 2 μL of fatty acid methyl esters (FAME) as a retention time index and 45 μL of N-methyl-N-trimethylsilyltrifluoroacetamide (MSTFA +1% TMCS; Thermo, USA) were added into the derivative and reacted for 60 min at 800rpm at 37°C.…”
