Integrin β3/Akt signaling contributes to platelet-induced hemangioendothelioma growth

Abstract: Hemangioendothelioma (HE) is a type of angiomatous lesions that features endothelial cell proliferation. Understanding the mechanisms orchestrating HE angiogenesis can provide therapeutic insights. It has been shown that platelets can support normal and malignant endothelial cells during angiogenesis. Using the mouse endothelial-derived EOMA cell line as a model of HE, we explored the regulatory effect of platelets. We found that platelets stimulated EOMA proliferation but did not mitigate apoptosis. Furthermo… Show more

“…To generate hemangioendothelioma, the injection of MS1 cells in mice was performed as previously described with minor modifications . Briefly, MS1 cells were collected, resuspended in DMEM medium (4.5 × 10 5 cells in 200 μL) and subjected to dorsally subcutaneous injection in nude mice.…”

“…P-selectin, a C-type lectin expressed on the surface of platelets, interacts with P-selectin ligand 1 (PSGL-1) and other mucins on cancer cells, while another mucinous glycoprotein, podoplanin (PDPN), which is upregulated in various types of cancer, can bind to the CLEC-2 receptor on platelets and induce TCIPA [11] . Furthermore, direct contact via platelet membrane molecules CLEC-2 or integrin β3 also promotes tumor growth in vitro and in vivo [ 19 , 20 ]. By providing pro-coagulant surface, releasing bioactive molecules, and interacting with leukocytes, CEPs contribute to TME and cancer-associated immunothrombosis, which further supports cancer progression [21] .…”

“…TGFβ1 can reduce the expression of E-cadherin (E-cad) and induce epithelial-mesenchymal transition (EMT) (19,20). The integrin β3 (ITGβ3)/AKT signaling pathway can promote the growth of platelet-induced hemangioendothelioma (21). Ni et al determined that the mechanism of CBX7 underlying its inhibitory effect on cell growth may involve the AKT signaling pathway (11).…”
