Interaction between physical exercise and APOE gene polymorphism on cognitive function in older people

Abstract: We aimed to present an overview of the literature regarding the interaction between physical exercise and APOE gene polymorphism on cognitive function, particularly in patients with Alzheimer's disease (AD). Firstly, this review focused on the effect of the physical exercise on cognitive function, regardless of APOE gene polymorphism. Some studies have shown that a high level of cardiorespiratory fitness is associated with less neuronal damage with an improvement in memory score tests whereas other studies fai… Show more

“…Importantly, the epigenetic regulation of gene expression mediated by histone deacetylases (HDACs), which act by removing acetyl groups from histones, resulting in gene silencing, has been demonstrated to play a substantial role in AD pathophysiology. , HDACs are critically involved in the regulation of the expression of genes involved in lipid metabolism and long-term potentiation, and HDAC inhibitors (HDACi’s) represent a promising therapeutic approach against AD . In this regard, pan class I HDACi’s were recently indicated to increase astrocytic APOE expression and secretion .…”

“…Physical exercise has been described as a protective factor against cognitive decline and AD. The effects of physical exercise can be modulated by genetic factors, as suggested in AD patients, where APOE4 allele carriers were more responsive to the beneficial effects of physical activity compared to non-carriers [ 84 ].…”

“…Regular physical activity also improved executive function, working memory, and cognitive flexibility [19]. However, such advantages of physical activity on cognition in people with AD were not universal across studies (reviewed in [20]), probably owing to heterogeneous samples and methodologies, and the lack of standardized protocols.…”
