Interactions Among Heparin, Cold-Insoluble Globulin, and Fibrinogen in Formation of the Heparin Precipitable Fraction of Plasma

Abstract: Fibrinogen and the cold-insoluble globulin of plasma (CIg) are the main protein components of the heparin cryoprecipi table fraction (HPF) of normal plasma. The interactions between these proteins and heparin were examined. Heparin formed a cold precipitable complex with purified CIg or with mixtures of CIg and fibrinogen (T/2, 0.2; pH 7.2) but not with fibrinogen alone. Cryoprecipitation could be augmented by addition of Ca++ or by selection of optimal heparin levels; it could be reduced or even abolished by … Show more

“…Ce complexe est toujours composé de fibrinogène, de fibronectine et de fibrine. Il peut également contenir de l'albumine, de l'␣1-antitrypsine, de l'␣2-macroglobuline et/ou une ou plusieurs immunoglobulines [8][9][10][11]. La fibronectine agit comme un noyau autour duquel vont se lier les complexes de fibrine-fibrinogène [12].…”

“…FBS was passed through a heparin-agarose column to remove fibronectin and laminin [30][31][32]. Approximately 10 ml of heparin-agarose gel was packed in a plastic column, 10 mm in diameter and 100 mm in length, and kept at 4 0 C. The gels were washed with PBS prior to use.…”

“…Since fibronectin and laminin are known to bind to heparin [30][31][32], we subjected FBS to heparin affinity chromatography to prepare fibronectin-and laminin-poor FBS. We then investigated the effects of a fibronectin-coated substratum upon the proliferation of trophoblast and EPC cells derived from mouse blastocysts cultured in vitro.…”
