Interferon Induction by Viruses and Polynucleotides: A Differential Effect of Camptothecin

Abstract: SUMMARYThe plant alkaloid camptothecin inhibits interferon production induced by Newcastle disease virus (NDV) or ultraviolet-irradiated NDV in chick and human cells, and by Sindbis virus in chick cells. It has no effect on interferon production induced by polyffI), poly(rC) in chick and human cells. No effect of camptothecin could be detected on the multiplication of NDV, and it is concluded that the inhibition reflects a difference between interferon induction by viruses and by polynucleotides.

“…Interferon was assayed by the RNA-reduction method (Atherton & Burke, 1975) with HFF cells for human fibroblast interferon and BT cells for lymphoblastoid interferon. The results are expressed in reference units, based on the human leukocyte interferon research standard (69/19), which is defined as containing 5ooo reference research units and which has a titre of/5oo in HFF cells and 5oooo in BT cells.…”

“…Samples for interferon assays were dialysed for 5 days at 4 °C against pH 2 buffer to inactivate residual virus. Interferon assays were carried out in human foreskin fibroblasts (HFF) (Atherton & Burke, 1975). Interferon titres are expressed as international units (IU): the interferon-fl international reference preparation (G-023 902 527) containing 10000 IU/ml, titrated at 5000 U/ml in HFF.…”
