Interferon-resistant Daudi Cell Line with a Stat2 Defect Is Resistant to Apoptosis Induced by Chemotherapeutic Agents

Abstract: Interferon-␣ (IFN␣) has shown promise in the treatment of various cancers. However, the development of IFN resistance is a significant drawback. Using conditions that mimic in vivo selection of IFN-resistant cells, the RST2 IFN-resistant cell line was isolated from the highly IFN-sensitive Daudi human Burkitt lymphoma cell line. The RST2 cell line was resistant to the antiviral, antiproliferative, and gene-induction actions of IFN␣. Although STAT2 mRNA was present, STAT2 protein expression was deficient in RST… Show more

“…Resistance to IFN can arise from defects in the IFN signaling pathway or from alterations in the apoptotic circuits. Defects in IRF-9 and STAT2 expression or epigenetic changes are responsible for resistance to IFNmediated anti-tumor activity and for apoptotic insensitivity [29][30][31][32][33]. On the other side, up-regulation of anti-apoptotic Bcl-2 family members and activation of the NF-kB pathway can also confer resistance to IFN-mediated cell death [34][35][36].…”

“…The induction of apoptosis was monitored by immunoblotting whole cell extracts for caspase-dependent PARP cleavage as previously described [14], or flow cytometry (Accuri Model 6C) using the Annexin V-FITC apoptosis detection kit (BD Pharmingen), according to the manufacturer’s instructions.…”

“…Du et al . discovered that IFN resistant cells produce a STAT2 splice variant containing intron 19 (40). This retained intron introduces a stop codon before the Src homology 2 domain, leading to disruption of STAT dimerization.…”
