Interleukin‐21 promotes osteoclastogenesis in humans with rheumatoid arthritis and in mice with collagen‐induced arthritis

Abstract: Objective. Bone destruction is a critical pathology involved in the functional disability caused by rheumatoid arthritis (RA). Osteoclasts, which are specialized bone-resorbing cells regulated by cytokines such as RANKL, are implicated in bone destruction in RA. The aim of this study was to determine whether interleukin-21 (IL-21), a potent immunomodulatory 4-␣-helical bundle type 1 cytokine, has osteoclastogenic activity in patients with RA and in mice with collageninduced arthritis (CIA).Methods. The express… Show more

“…Taken together, the results from both in vitro and in vivo experiments suggested evaluating effects of Il-21 in human CNS autoimmune demyelinating disease. It has been reported that serum Il-21 levels were elevated in many autoimmune diseases, such as systemic lupus erythematosus, rheumatoid arthritis and Sjogren's syndrome 30,31 . Consistent with previous findings, we demonstrated a high level of serum Il-21 in NMO THe CANADIAN JOURNAl OF NeUROlOGICAl SCIeNCeS 818…”

“…IL-21, a new member of the type 1 cytokine superfamily, promotes osteoclastogenesis in RA (Ref. 215) and has been suggested as target for immune-mediated diseases, especially for preventing bone destruction (Ref. As discussed throughout this review, research into the molecular pathogenesis of periodontitis is continuously producing novel and significant results.…”

“…IL-7 induces overexpression of RANKL in T lymphocytes and its blockade significantly reduces monocyte attraction, osteoclast differentiation and bone erosion in CIA [71]. IL-21 stimulates osteoclastogenesis through an induction of RANKL in CD4 + T lymphocytes, and is upregulated in the synovia and sera of RA patients [72]. Although its role in osteoclastogenesis is still controversial, IL-18 is known to induce M-CSF, GM-CSF, RANKL and OPG in synovial fibroblasts and/or T lymphocytes [73].…”
