Interleukin-6–Deficient Mice Resist Development of Autoimmune Myocarditis Associated With Impaired Upregulation of Complement C3

Abstract: Background-Interleukin (IL)-6 regulates various aspects of the immune response. In the context of heart diseases, it has been recognized as a prognostic factor for dilated cardiomyopathy, which often results from myocarditis. Methods and Results-Using IL-6 -deficient mice, we studied the role of IL-6 in a model of autoimmune myocarditis resulting from immunization with a peptide derived from cardiac ␣-myosin. Prevalence and severity of myocarditis were markedly reduced in the absence of IL-6. CD4 ϩ T cells fro… Show more

“…In this context, it has been shown recently, that IL-6, which is essential for the induction of Th17 T cells (8), and IL-23-promoting Th17 expansion are both critical for EAM development (12,13). Our data prove the direct pathogenic role of IL-17 in EAM development, because IL-17 depletion prevents EAM development.…”

“…Mice were immunized with 150 g of the MyHC-␣ (MyHC-␣ 614 -634 ) Ac-SLKLMATLFSTYASAD-OH emulsified 1:1 in PBS/CFA (1 mg/ml, H37Ra; Difco) as described (8).…”

“…Mice were inoculated with 100 mg of MyHC-a (MyHC-a 614-629; Ac-SLKLMATLFSTYASAD-OH), emulsified at a 1:1 ratio in PBS/CFA at days 0 and 7 as previously described [48]. At different time points, the mice were anaesthetized with pento- Figure 6.…”

“…A number of proinflammatory cytokines, including IL-1b, IL-6, IL-12, TNF-a, and GM-CSF, have been shown to contribute to the development of autoimmune myocarditis in animal models and human cases (8)(9)(10)(11)(12)(13). EAM is a CD4 + T cell-mediated disease (7,14), and activation of self-Ag-loaded dendritic cells (DCs) is critical for expansion of autoreactive CD4 + T cells.…”
