Intermediate Lobe Pituitary Hormone

Abstract: 1. The optimum source for the manufacture of melanophore hormone (B hormone) is the posterior lobe of the pituitary, provided that the pressor and oxytocic autacoids can be removed without recourse to caustic soda treatment. 2. Previous methods of assaying melanophore activity are criticised, and the advantages of hypophysectomised Xenopus lævis as a test animal are emphasised. 3. A provisional unit of melanophore activity is proposed. 4. A new and rapid method is described for the preparation of B extracts wh… Show more

“…By definition, 1 ml of extract of International Standard Posterior Lobe Powder contains 2-0 i.u. pressor, antidiuretic, oxytocic and melano¬ phore-expanding activity (Landgrebe & Waring, 1941). Preparations of pure melanophore-expanding polypeptide from pig pituitaries contain 1500 i.u.…”

“…What evidence there is makes (c) the most probable for the following reasons. Xenopus injected with similar doses of caustic soda treated and untreated extracts of comparable purity show only barely significant differences in the speed of initial response [see curves in Landgrebe and Waring, 1941]. So it is unlikely that the difference between prolonged and short responses is due to difference in the speed that the extracts diffuse through membranes (i.e.…”

“…There have been many claims for other pharmacological actions of "melanophore expanding hormone" [see Landgrebe and Waring, 1941], but most of these are invalidated because authors have not specified both the melanophore expanding potency and the exact method of preparation of their extracts. This should be done for all glandular extracts containing protein.…”
