A CE method for metacycline (MTC) determination was investigated in an inter-laboratory experiment. Many problems were encountered in this study, most of which were related to the transfer of the method to different CE equipment. The reported problems could be classified into different categories: problems related to the precision, to the parameters in the protocol, and to the MTC peak shape. As the peak shape problem was partially responsible for the poor precision, a new CE method was developed in order to obtain a good MTC peak shape on all equipment. The precision of this new method for MTC determination was examined in an intermediate precision study, where the influence of the factors "time" and "equipment" was investigated. Although the new method could be transferred to different instruments, the precision remained poor mainly due to the contributions of the between-replicate and the between-injection variances.

Drug monitoring in serum samples was performed using second-order data generated by CE-DAD, processed with a suitable chemometric strategy. Carbamazepine could be accurately quantitated in the presence of its main metabolite (carbamazepine epoxide), other therapeutic drugs (lamotrigine, phenobarbital, phenytoin, phenylephrine, ibuprofen, acetaminophen, theophylline, caffeine, acetyl salicylic acid), and additional serum endogenous components. The analytical strategy consisted of the following steps: (i) serum sample clean-up to remove matrix interferences, (ii) data pre-processing, in order to reduce the background and to correct for electrophoretic time shifts, and (iii) resolution of fully overlapped CE peaks (corresponding to carbamazepine, its metabolite, lamotrigine and unexpected serum components) by the well-known multivariate curve resolution-alternating least squares algorithm, which extracts quantitative information that can be uniquely ascribed to the analyte of interest. The analyte concentration in serum samples ranged from 2.00 to 8.00 mg/L. Mean recoveries were 102.6% (s=7.7) for binary samples, and 94.8% (s=13.5) for spiked serum samples, while CV (%)=4.0 was computed for five replicate, indicative of the acceptable accuracy and precision of the proposed method.

Because of the problems with stability and solubility, some of the amino acids (AAs) used in parenteral nutrition products are often replaced by their acetylated forms. These include N-acetyltyrosine (NAT) and N-acetylcysteine (NAC). This leads to a need to develop new analytical methods for rapid and easy determination of these substances in the presence of common AAs. Capillary electrophoresis (CE) is one of the techniques that have been successfully applied to the assay of multi-component samples containing AAs and their derivatives. This paper discusses a new CE method for the simultaneous determination of two acetylated AAs in solutions for parenteral nutrition. A background electrolyte (BGE) was developed based on borate buffer with alkaline pH. The method is selective and enables the separation and assay of analytes without special sample pretreatment. Validation parameters confirmed sufficient precision and accuracy of the method. Its applicability was verified by testing several medicinal products from various manufacturers. Moreover, the flexibility of the method was checked using two brands of CE equipment. Appropriate adjustment of instrumental parameters turned out to be essential for method transferability. The method could be used in routine testing of parenteral nutrition medicinal products containing AAs.
