Intracellular interleukin (IL)-1 family cytokine processing enzyme

Abstract: The interleukin (IL)-1 family is the largest family of interleukins. Eleven members of the IL-1 family of ligands are intracellular molecules, except a single isoform of an IL-1 receptor antagonist (IL-1Ra; also known as IL-1RN), which contains a signal peptide at the N-terminus for effective secretion. The inflammasome is a complex of intracellular molecules that is responsible for the processing of IL-1β and IL-18, whereas the remaining IL-1 family members, including IL-1α, are processed in an inflammasome c… Show more

“…Their receptors share the same ligand binding chain and signal transducing chain. However, different from IL‐1β, IL‐1α activity is independent of the inflammasome caspase‐1 pathway. Regarding inflammation‐induced arthritis and cartilage degeneration, IL‐1β has been mentioned considerably more than IL‐1α.…”

“…The biogenesis of most IL-1 family members differs from other cytokines insofar as most IL-1 interleukins are generated as biologically inactive zymogen forms which are proteolytically cleaved [54]. In the case of IL-1β (and IL-18), N-terminal processing is facilitated in myeloid cell types by caspases which often serve as the effector component of inflammasomes.…”

“…IL-1α and IL-1β are the main subtypes of the IL-1 family 8 . IL-1α has similar effects to IL-1β by binding to the IL-1 receptor 1 (IL-1R).…”
