Intracellular melatonin distribution in cultured cell lines

Abstract: A specific antibody combined with a fluorescein-labeled immunoglobulin was used to investigate the topographic distribution of melatonin in a variety of cells of different origins. Positive identification of both nuclear and cytosolic melatonin was confirmed in all the tested cells: Swiss 3T3 mouse fibroblasts, BCG1 bovine granulosa, NB41A3 mouse neuroblastoma, F9 mouse teratocarcinoma, MDCK normal canine kidney derived and human HeLa cell lines, as well as in human peripheral blood mononuclear leukocytes and … Show more

“…19,24 Longterm (5 days) BrdU incorporation into 1-day-old LM3 and B16 spheroids yielded intense BrdU nuclear staining distributed throughout the spheroid (Fig 4a-c). On the other hand, with short-lasting BrdU pulses (1 day) into 5-day-old MCS, there was a high fraction of nonproliferating or quiescent cells (Fig 4d-f).…”

“…19 At the end of the incorporation period, the medium was removed, and MCS were washed twice in PBS and resuspended in Bouin for 20 minutes. After washing three times for 5 minutes each in PBS, the pellet was resuspended in 2% liquid low-melting point agar in PBS, dehydrated in ethanol, embedded in paraffin and sectioned.…”

“…The endogenous production of this molecule falls dramatically with age (Djeridane et al 2005) coincidental with the onset of many of the age-associated neurodegenerative conditions (Hurn et al 1996;Karasek 2004); 2. Melatonin readily crosses the blood brain barrier and after its exogenous administration it is found in high concentrations in the brain, sometimes exceeding those in the blood manifold (Mené ndez-Pelá ez Finnochiaro and Glikin 1998); …”

“…At the end of the 1980s, the works of Finocchiaro suggested that human peripheral blood mononuclear cells (PBMCs) possessed the capacity to produce melatonin in vitro after being cultured in the presence of IFN-γ or serotonin [36,37]. In early 2000, two studies revealed the presence of high concentrations of melatonin in rat, mouse and human bone marrow [38,39].…”
