Intracellular transport dynamics revealed by single-particle tracking

Abstract: Intracellular transport is the basis for the transfer of matter, energy, and information in cells and is critical to many cellular functions. Within the nonequilibrium environment of living cells, the transport behaviours are far from the traditional motion in liquid but are more complex and active. With the advantage of high spatial and temporal resolution, the single-particle tracking (SPT) method is widely utilized and has achieved great advances in revealing intracellular transport dynamics. “…ApoE-ALPs could be developed as a promising drug delivery for treating tumor types overexpressing LDLR. In addition, many nanoparticles have indeterminant uptake pathways due to heterogeneity of their shape, size, and surface chemistry, which hinders their efficient delivery ( 42 , 43 ). However, our results demonstrate that ALPs are internalized through a defined and predictable pathway.…”

“…In contrast, the Eulerian methods are faster, as they calculate ensemble statistics, with the drawback that subtle differences between individual particles will not be noticed. SPT has been applied in a diverse set of biological contexts, including dynamics of membrane receptors ( 104 , 108 , 156 ), actin dynamics ( 2 , 50 , 189 ), intracellular transport dynamics ( 127 , 129 , 191 ), and transcription factor mobility in the nucleus ( 78 , 93 , 167 , 188 ).…”

“…Intracellular transport of various cargos in the microtubule network is driven by molecular motors with the energy generated from ATP hydrolysis. , The characteristic translational and rotational motions of cargos can be highly informative on the binding states between cargos and motor proteins and offer a new angle to elucidate the cargo–motor–microtubule interactions.…”
