Intracerebroventricular infusion of cytosine-arabinoside causes prepulse inhibition disruption

Abstract: Adult neurogenesis in hippocampus is associated with behaviors such as learning. Hippocampus is involved in the regulation of prepulse inhibition (PPI), but the relationship between neurogenesis and PPI is unexplored. We conducted four experiments to determine the role of neural progenitor cell proliferation in PPI. Intracerebroventricular infusion of cytostatic cytosine arabinoside caused PPI disruption but repeated exposure to PPI sessions prevented the PPI disruption. Corticosterone treatment, which decreas… Show more

“…The Ara-c group received oral feeding of 1 mg/kg LBP and infusion of cytosine arabinoside (Ara-c, a cytostatic drug which blocks cell proliferation) for seven days. Procedures of infusion was described previously [21]. In brief, osmotic pumps (Alzet, California, USA) filled with saline/Ara-c were implanted subcutaneously at the dorsal neck region of the rats.…”

“…Prepulse inhibition of the acoustic startle response is consistently unaltered by wheel running (Lau et al, 2009; Pietropaolo et al, 2006; Salam et al, 2009). In line with the idea that a history of wheel running produces beneficial effects on startle, mice that ran on a wheel for 2 wks exhibited reduced acoustic startle amplitude relative to sedentary counterparts (Salam et al, 2009).…”

“…One is to ablate neurogenesis at the time of injury to determine whether this then impairs recovery. Strategies to do this include systemic or local administration of anti-mitotic agents such as Ara-C that are known to affect dividing neural progenitors (Doetsch et al, 1999; Lau et al, 2009). Another is to perform cranial irradiation directed to neurogenic zones such as the SVZ and dentate gyrus in order to more selectively impair neurogenesis (Hellstrom et al, 2008; Naylor et al, 2008).…”
